A study was undertaken on three species of fresh fishes which have an im-portant role to develop a fast and simple method for extraction. An efficient, practical and reproducible method was used in this study. Wet tissues were homogenized with mixtures of chloroform and methanol in several proportions of solvent and water from tissues. The dilution with chloro-form and water separated the homogenate with two layers. The chloroform layer containing all lipids and the methanol layer all non lipid component. The pure lipid extraction was obtained of the chloroform layer. The result achieved from this analysis are as follows: Tuna fish contains 0,0425 % chlolesterol; Commond Carp contains 0,0315 % chlolesterol; Milkfish contains 0,02475 % chlolesterol. The cholesterol content was determined by a visible light spectrophotometer on 630 nm wave length.
